Actually, given that the red Mafia had a Burden with them I wouldn't put it past the hosts to give them a 5th member, maybe a regular Mafia or that one Mafia who's kill only succeed at 50% all the times. If they really wanted to push for a Mafia win, then that would be the way to go.

It doesn't matter. The relic description says they get an extra kill even if they used up their kill already. Likely worded like that just for the assassin case. And you can bet they are firmly holding onto the relic since day 2.
