Question A few general questions regarding the plot

So I've just finished watching the english sub episodes and have now begun reading the manga which follows. But the series has me left me wondering a few things.
One question would be are the titan shifters born with this power to shift? If so what was Eren's injection for?
Secondly, where did the other 4 titan shifters come from? 3 of whom seem to share a history together.
Last of all, what's with the titan shifters trying to eradicate humanity?

All answers would be greatly appreciated! Thanks!

The manga continues monthly. If you have only seen the anime , I suggest you also read the manga for even more mind blowing events. If you are already reading the manga, I suggest you keep going. In any case, welcome to the forums.

Unknown:
Maybe the are born with it and only certain children with the right DNA
can be turned into ones by injection them with the serum.
It might only work at a certain age, teenage years would be probable in this case.
Another scenario would be that the injection had an unlocking affect
to his memories - we don't know if Eren is the actual son of Grisha
he could have found and adopted him, shifters don't age while transformed.
There is also the special ability of the coordinate that he might have
got from the serum or it did both making him a titan with special ability.

Reiner, Annie & Berthold come from a yet to be revealed "village" outside
the walls Ymir:

Was a mindless titan roaming outside the walls for 60 years
who ate one of their friends and thus by eating a shifter was enabled
to switch between titan mode and being a human she then used the
confusion caused by RABs attacked to slip inside the Walls...

The don't want to eradicate humanity - there are humans themselfs but
they were willing to destroy the wall-civilization but abandoned that plan
after the found out about Eren.
Why are the doing it?
There is no definte answer to that but according to RABY the days
inside the walls are numbered - Hell was a word used about it's future...
RAB probably considered them dead or better of dead when the attacked.

Yeah I do remember reading a hypothesis that Erne is in fact not grishas son, and that what he injected him with is the unknown coordinate, which is why annie was so desperate to capture him alive. What is the significance and origin of the coordinate ?

But I thought it was discovered by hanji that the government want eren so that lord reiss could eat him and get his shifter/coordinate powers... Then your hypothesis slightly makes sense seen as the government have taken little interest in the other shifters!

What is the significance and origin of the coordinate

It's so important that RAB abandoned their original plan in favor of it
if it wasn't the thing the wanted all along...
It's so important that it could safe the Wall civilization from a dystopian future
according to Reiner - but he might just said that to persuade Eren.
The only time we saw it's application was when Eren used
it to get some mindless titans to attack an abnormal one in a
moment of desperation and furry...
And from that single time and some mentions of deleted/locked away memories
we can argue in what ways that power can be used to manipulate
titans and humans.

RAB acted for a power outside of the walls considering how the infiltrated
and tried to steal something the goverment wants makes it rather clear.

But someone inside the walls might have known about the attack since
Grisha was moving to the inner walls not long before it.
Since he left his family behind he himself likely did not know.
