Actually we don't really how the "Demon marks" works but if I had to guess I'll say it's a switch button to use your Darkness.
Activating your mark means you can use/deploy your Darkness and when the mark grow (2e mark, AM,...) it mean you're tapping into more Darkness.
It's just a guess but when Derrieri fought Denzel/Nerobosta she was angry and made her mark grow, and with that the Darkness in her arm had tenfold.
And with his AM, Meliodas deploy his Darkness to cover himself, so his mark widens all over his forehead

And how many demon marks does meliodas have?

Every Demon has only one Demon mark and it's the same for Meliodas. He, Zeldoris and the Demon King have the same Demon mark (the spiral thing) but Meliodas, as far as we know for now, can tap on more Darkness then any other demon (except the DK ofc). That's why he can grow his mark til it widens all over his forehead.
You can notice the change in others Demons too. For exemple, when they turn into their battle form, the mark of the Masters grow

As far as meliodas goes, I believe we have seen 4 or 5 demon marks. First is the original demon mark which he shares with his father and brother. Second would be the one he had against dolor and gloxinia, the one that passes over his eye. Then the one that is on the center of his forehead. But to me this one is kinda tricky because it does not seem to be drawn consistently. We have seen versions of this one that don't make a full circle and sometimes it is a full circle with a little spot in the middle. And then finally the assault mode.
